DESCRIPTION:Objectives:\n– Acquire the theoretical foundations of sample preparation and 3D SEM imaging. \n– Acquire the practical skills and tools needed to prepare biological samples involving high-pressure fixation and rapid cryo-substitution for 3D SEM imaging. \n– Attend SEM3D imaging acquisition on a prepared sample\, and acquire the skills to align images of a volume after acquisition using FIJI software. \nAt the end of the course\, trainees will be able to : \n– define a sample preparation strategy to answer a biological question requiring 3D SEM \n– Carry out an A to Z protocol for rapid cryosubstitution (QFS) \n– Align a 3D SEM stack
