Training workshop on 3D SEM techniques, from sample preparation to image acquisition

Objectives:
– Acquire the theoretical foundations of sample preparation and 3D SEM imaging.

– Acquire the practical skills and tools needed to prepare biological samples involving high-pressure fixation and rapid cryo-substitution for 3D SEM imaging.

– Attend SEM3D imaging acquisition on a prepared sample, and acquire the skills to align images of a volume after acquisition using FIJI software.

At the end of the course, trainees will be able to :

– define a sample preparation strategy to answer a biological question requiring 3D SEM

– Carry out an A to Z protocol for rapid cryosubstitution (QFS)

– Align a 3D SEM stack
